Abstract
The key bottleneck for the continued success of precision medicine in cancer lies in identifying more targetable genes and associated efficacious clinically usable inhibitors. In this issue of <i>Cancer Research</i>, Hamanaka and colleagues identify YES1 kinase as a targetable cancer target and generate an effective chemical inhibitor for YES1 and demonstrate its efficacy in YES1-amplified tumors.<i>See related article by Hamanaka et al., p. 5734</i>.
