Abstract
HER2-directed tyrosine kinase inhibitor combinations with trastuzumab represent an emerging strategy for hormone receptor-negative or -positive metastatic breast cancer with activating HER2 mutations. Correlative genomics reveal on-target and downstream molecular evolution during treatment, identifying response patterns and potential acquired resistance mechanisms. Refining precision therapies for HER2-mutant breast cancer remains a priority.
